Release is a quality unit decision, backed by records

In the US, the quality control unit has “responsibility and authority to approve or reject all components, drug product containers, closures, in-process materials, packaging material, labeling, and drug products” (21 CFR 211.22(a)). In FY2025, § 211.22(d), which requires those responsibilities and procedures to be in writing and followed, was FDA’s most-cited drug observation, with 243 citations (GMP Insiders).

US requirements before release

Requirement21 CFR Part 211
Lab testing”For each batch of drug product, there shall be appropriate laboratory determination of satisfactory conformance to final specifications … prior to release” (§ 211.165(a))
Complete batch record”complete information relating to the production and control of each batch,” including dates, equipment, component lots, weights, in-process and lab results, line clearance, yield, labeling specimens, sampling, and who performed and checked each step (§ 211.188(b))
Record reviewRecords “shall be reviewed and approved by the quality control unit … before a batch is released or distributed” (§ 211.192)
DiscrepanciesInvestigated “whether or not the batch has already been distributed” (§ 211.192)

EU requirements: QP certification under Annex 16

In the EU, “Each batch of finished product must be certified by a QP within the EU before being released” (EU GMP Annex 16). Annex 16 lists the points the QP must ensure, starting with confirmation that “All sites of manufacture, analysis and certification are compliant with the terms of the MA for the intended territory,” and requires certification records kept “for at least five years.” US companies exporting to the EU need their documentation to support that certification.

  • Faster, exception-based review. Review by exception, endorsed in GAMP guidance, can shrink a 150-page record review to a short exception report (EY; ISPE).
  • Onshoring. In August 2025, FDA announced PreCheck, a two-phase program to support new US manufacturing facilities (Sidley). New sites will need release processes that work from day one.
  • EU documentation rules are changing. The 2025 draft revision of GMP Chapter 4 puts risk management at the center of data governance (European Commission). It is still a draft.

Batch release checklist

Batch record

  • Master record version matches the approved, current version
  • All steps completed, signed and verified by a second person where required
  • Component and material lots recorded and released
  • Equipment used is identified, cleaned and within calibration
  • In-process results within limits
  • Actual yield and percentage of theoretical yield recorded and within limits
  • Labeling specimens attached and reconciled; line clearance documented
  • Corrections follow good documentation practice

Data and audit trails

  • Electronic records and relevant audit trails reviewed before approval
  • No unexplained data changes, deletions or aborted runs

Laboratory

  • All release tests complete and conforming to specifications
  • OOS or OOT results investigated and closed
  • Certificates of analysis verified against specifications

Deviations and changes

  • All deviations for the batch closed, or impact assessed and justified for release
  • Change controls affecting the batch approved and implemented
  • Investigation scope extended to other affected batches where needed

Regulatory

  • Manufacture complies with the approved application (NDA/ANDA/BLA) or EU marketing authorization
  • For EU release: QP certification points under Annex 16 confirmed and recorded in the register

Decision

  • Quality unit approval documented with date and signature
  • Batch status updated in inventory systems

Speed up release without cutting corners

Release delays usually come from searching: for the open deviation, the missing signature, the audit trail entry no one reviewed. AI document assessment can review batch records and supporting documents against your release checklist, flag exceptions and cite the page behind every finding, so QA reviews exceptions instead of every page.

Run a sample regulatory document assessment on a closed batch record. AskDona shows the evidence; your QA team makes the release decision.

FAQ

Who can release a batch in the US? The quality control unit, which has authority to approve or reject drug products (21 CFR 211.22).

What must be reviewed before release? All production and control records, including lab results and investigations, under § 211.192 and § 211.165.

What is QP certification? In the EU, a Qualified Person must certify each batch before release, per Annex 16 (EU GMP Annex 16).

Can AI release a batch? No. AI can prepare and prioritize the review; the quality unit or QP makes the decision.
